Involvement of Microsomal Na/K ATPase Activity in the Mechanism of Action of Dopaminergic D2 Receptors

چکیده
Problem statement: Our previous research has shown that mitochondrial ATPase may have a role in the action of dopaminergic D2 receptors. It is therefore, thought of interest to investigate the effect of bromocriptine, sulpiride or their combination on the mitochondrial and microsomal ouabain sensitive and ouabain insensitive Na/K-ATPase in the liver of rats. Approach: In vivo treatment was carried out to study the effects of bromocriptine mesylate 10 mg kg, sulpiride 10 mg kg; or bromocriptine mesylate 10 mg kg + sulpiride 10 mg kg, compared to a control group, in order to measure liver Na/K-ATPase activity. The drugs were injected intraperitoneally daily for 14 days. Mitochondrial and microsomal fractions were obtained by the methods of Schwartz. Results: Pretreatment of rats with both bromocriptine and sulpiride given simultaneously resulted in complete abolition of stimulation caused by bromocriptine and inhibition that resulted from sulpiride on mitochondrial as well as microsomal ATPase activities. Conclusion: This study indicated that the in vivo administration of dopaminergic agonist and antagonist drugs result in a significant alteration not only in the mitochondrial ATPase enzyme activity but also in the microsomal ATPase enzyme activity. Therefore, it is quite possible that ATPase of the tow fractions play important physiological as well as pathological roles in the mechanism of action of dopaminergic receptors.

The Involvement of Intra-Hippocampal Dopamine Receptors in the Conditioned Place Preference Induced By Orexin Administration into the Rat Ventral Tegmental Area
The activity of dopamine (DA)-containing neurons in the ventral tegmental area (VTA) is a key mechanism in mesolimbic reward processing that has modulatory effects on different diencephalic structures like hippocampus (HIP), and receives inhibitory feedback and excitatory feed forward control. In addition, within the hippocampus, DA receptors are mostly located in the dorsal part (CA1) and dopa...

متن کاملO-10: A Marked Animal-Vegetal Polarity in The Localization of Na+,K+-ATPase Activity and Its Down-Regulation Following Progesterone-Induced Maturation
Background: Polarized cells are key to the process of differentiation. Xenopus oocyte is a polarized cell that has complete blue-print to differentiate 3 germ layers following fertilization, as key determinant molecules (Proteins and RNAs) are asymmetrically localized. The objective of this work was to localize Na+, K+-ATPase activity along animal-vegetal axis of polarized Xenopus oocyte and fo...
متن کاملPossible Involvement of Glutamatergic, Adrenergic and Dopaminergic System in Methylphenidate - induced Motor Activity and Mood-related Alterations in Rats
Background and Objective: Methylphenidate (MPH), as a central nervous system stimulant, is often used to manage hyperactive disorders. The literature is scarce regarding the behavioral consequences of chronic MPH treatment and the role of involved receptors. Thus, in the current study involved receptors in MPH induced-anxiety, depression and motor activity disorders were evaluated. Materials a...
متن کاملO-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells
Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...
